COURSE DESCRIPTION
Prerequisites: (W) (M), BIO 2125.
1 CREDIT. The student will earn to successfully
maintain tissue culture cells in a healthy uncontaminated state for an
extended period of time. The course will include making tissue culture
media, monitoring cell growth, freezing cells, and bringing up frozen
cells.
COURSE PACING
BIOT 2922 is a combination of a group-paced and individually-paced course.
Your instructor will establish objectives to be met within the time parameters
of the semester. These dates must be observed in order to maintain good
standing in the class.
COURSE GRADING CRITERIA
A: 90-100% average
B: 80-89% average
C: 70-79% average
D: 65-69% average
F: Below 65% average
Grades will be based on your level of mastery of the course objectives.

professional judgment of Biotechnology faculty and upon comparable courses
from other institutions of higher education.
ATTENDANCE POLICY:
You are expected to attend and actively participate in all scheduled classes
for this course.

Tissue Culture
1. The student learns to make tissue culture media: to filter-sterilize
components, combine, adjust pH and then to test it for sterility.
2. The student learns to monitor cell growth by observations of tissue
culture medium changes and by observations of cells with inverted phase
microscope.
3. The student learns to use the hemocytometer to count viable cells,
and will perform counts at least three times.
4. The student learns to calculate cell concentrations from cell counts,
and to calculate amounts to transfer to maintain optimum cell concentrations
for running stock and for freezing.
5. The student learns and uses aseptic transfer skills in laminar flow
hood .
6. The student learns and successfully performs procedures for freezing
down cells and bringing up cells from the frozen state.
7. The student learns to maintain tissue culture cells in healthy uncontaminated
state for a period of several weeks, using the skills listed above. The
student will maintain his/her cell line for at least a week in PSF-free
media without contamination
8. The student can distinguish between distilled and deionized water.
The student can list all the purification steps found in our water purification
system and describes briefly what each step accomplishes. The student
can explain what is meant by type I water, and why it cannot be stored
for any length of time. The student can describe how the quality of type
I water is monitored.
9. The student will perform the above tissue culture procedures on two
different cell lines: suspension mouse myeloma cells and adherent
CHO cells.
10. The student will maintain acceptable documentation of all manipulations,
observations, and procedures in a scientific notebook.
