Instructions:  Making Agarose for Gels

 

1.      Need recipe/protocol/directions.  (.8%, 1%, 2%, etc.)

 

2.      Use a beaker that is at least 2X-4X greater volume than the volume you are preparing.

 

3.      Follow recipe directions by adding correct amount of agarose powder to correct volume of buffer.

 

4.      Place plastic wrap over top and VENT- do NOT seal off beaker.

 

5.      Place beaker in microwave and set for about 1-1 ˝ minutes.

 

6.      Heat till bubbles start to form- WATCH that it doesn’t boil over.

 

7.      About every 30 seconds or so, remove beaker and carefully swirl solution.  (ALWAYS handle beaker with tongs or potholder.)

 

8.      Continue heating- stir/swirl- check that it is dissolved.

 

9.      When solution turns completely clear, it is done! 

 

10.  Cool to about 50 degrees C (comfortable handling temperature) before pouring.  Cool too much and it solidifies.  If too hot, it can warp the casting tray and parts.
